The sitter was the daughter of Don Pedro di Toledo, Viceroy of Naples, and in 1539 she married Cosimo de' Medici, 2nd Duke of Florence and later 1st Grand Duke of Tuscany. She was the mother of two succeeding Grand Dukes: Ferdinando I and Franceso. The latter married Bianca Capello, whom this portrait was formerly thought to represent.
